Even if Itsuki didn't do anything against Orochimaru, the latter would still come after him or his children.

Itsuki didn't want Pedomaru to scheme to take over any of his children's bodies as perfect containers.

Being strong Senjus his children would be the perfect research material for a person like Orochimaru.
